Hematological and biochemical effect of subcutaneous administration of robenacoxib in different goat breeds

Aim: This study aimed to investigate the effect of robenacoxib on hematological and biochemical parameters in healthy Alpine and Saanen goats 24 hours after subcutaneous administration. Materials and Methods: 12 healthy Alpine (n=6) and Saanen (n=6) breed goats were used. Robenacoxib was administered to Alpine and Saanen goats at a dose of 4 mg/kg once subcutaneously. Blood samples were collected before drug administration (0 hour) and then 24-hours. Hematological and biochemical parameters were analyzed using a hematology analyzer and biochemistry auto analyzer, respectively. Results: No change was observed in the clinical examination findings of both goat breeds. Statistical differences in hematological and biochemical parameters were determined within physiological limits in both goat breeds. The white blood cell, red blood cell and lymphocyte values were found to be low, and the monocyte% value was found to be high in Alpine goats. Decreases in red blood cell, hemoglobin and hematocrit levels and increases in the mean corpuscular volume and mean corpuscular hemoglobin values were observed at 24-hours in Saanen goats. Robenacoxib increases blood urea nitrogen value in both goat breeds, it decreases creatinine value in Saanen goats. Conclusion: There were no significant changes in hematological and biochemical parameters after robenacoxib administrations to Alpine and Saanen goats. Consequently, further studies are needed to investigate the effects of robenacoxib in high-dose and repeated administrations, liver and kidney dysfunctions, different age, breed, and physiological conditions. It was evaluated that robenacoxib could be used as an alternative NSAID to prevent pain and inflammation in goats, since no adverse effect was observed in the blood profile.

Evaluation of the Performance of Diagnostic Methods of Canine Parvovirus-2 and Canine Enteric Coronavirus Infections under Different Storage Conditions and Determination of Molecular Characterization

Aim: This research was carried out to detect CPV and CCoV infections in dogs in comparison with rapid kit and PCR and to determine the molecular characterization of these infections in Konya region. Besides, it was aimed to determine the sensitivity and specificity rates of the diagnostic tests after fresh or freeze-thawed stool for infection diagnosis. Materials and Methods: Faecal samples were collected from 50 unvaccinated, 0-12 months old dogs with diarrhoea symptoms at the shelter. The samples were analysed for CPV and CCoV by rapid test and PCR test. After freezethawing, the samples were checked again with the same tests. Results: CPV was positively diagnosed by rapid test and PCR in 2 and 29 fresh stool samples, respectively, and CCoV in 14 and 28 samples. CPV positive samples did not change while CCoV was diagnosed as positive in 10 samples and 28 samples by rapid test and PCR, respectively, after the freeze-thaw procedure. Although there were no differences in the diagnosis of CPV, the sensitivity of the rapid test in the diagnosis of CCoV decreased after the freezethaw procedure. In addition, only CPV-2b type was detected in CPV positive samples and both GI and GII subtypes were detected in CCoV positive samples as molecular. In conclusion, it was observed that rapid tests are not sensitive for accurate diagnosis of CPV and CCoV infections. Conclusion: The importance of choosing molecular diagnostic methods and using fresh samples for accurate diagnosis of virological infections can be emphasized.

Computed tomography reconstruction and morphometric analysis of the humerus and femur in New Zealand rabbits

Aim: This study was carried out to create three-dimensional (3D) models of the humerus and femur using computed tomography (CT) images of the New Zealand rabbit and to reveal whether there are differences between sexes and the right and left sides. Materials and Methods: The study was applied on 14 month old (10 male and 10 female) New Zealand rabbits. First, 0.5 mm thick images of the animals were taken by computed tomography (CT) and saved in DICOM format. Then, 3D models of the humerus and femur were obtained by reconstructing the images with the MIMICS 20.1 program. Morphometric data were obtained from the obtained 3D model. After this procedure, the rabbits were dissected and the weights of the humerus and femur were measured. Results: It was determined that the right humerus and femur volume and surface area in male rabbits were larger from the left side (p<0.05). There was no difference between the other morphometric values examined and rabbit bones did not show homotypic variation. In addition, it was thought that the humerus and femur bones could not be used to determine gender because the differences between the sexes were insignificant. Conclusion: It was thought that this study would provide the basis for studies and experimental models on rabbits by revealing the morphological features of the humerus and femur and would help the physician in the diagnosis of diseases by forming the basis of the data obtained with the imaging systems used in the clinical field.

Investigation of Nosema apis and Nosema ceranae in bees by Multiplex PCR in Kırıkkale Region

Aim: The aim of this study was conducted to determine the presence and prevalence of Nosemosis in honeybees in Kırıkkale region by Multiplex PCR. Materials and Methods: Samples were collected from 52 apiaries in Kırıkkale City Center, Bahşili, Balışeyh, Keskin, Sulakyurt and Yahşihan districts. The thorax and abdomen of ten bees collected from every 5 colonies were dissected and crushed with sterile physiological water in a porcelain mortar. The samples with Nosema spores were stained with Saffron and the multiplex PCR was applied to detect specimens to the same samples. Results: Nosema spp. spores were observed in 12 out of 52 apiaries (23.07%) microscopically. Among the positive samples, Balışeyh county was found with the highest rate of 57.14%. The township district is followed by Delice district with 33.33% and Kırıkkale /Center district with a ratio of 23.07%. Nosema spores were not found in samples taken from Bahşili, Keskin, Sulakyurt and Yahşihan districts. In general, the infection rates of the districts were determined as Central (5.76%), Balışeyh (7.69%), and Delice (9.61%). All the positive samples were detected as N. ceranae by multiplex PCR. Conclusion: This is the first study to determine the presence and prevalence of Nosemosis in honey bees in the Kırıkkale region and its etiological agent, N. ceranae. In line with the data obtained in this study, it was concluded that Nosemosis should be taken into consideration in bee losses in the region and beekeepers should be made aware of the issue.

Carvacrol alleviates acrylamide-induced hepatotoxicity in rats: An Experimental insights

Aim: This study aimed to investigate the protective effects of carvacrol (CRV) on rat liver against acrylamide (AA)-induced hepatotoxicity. Materials and Methods: A total of 30 Wistar Albino rats were used in the study. Rats were divided into 4 groups: Control (n:6), CRV (n:8, 50 mg/kg/ day carvacrol, i.p), AA (n:8, 40 mg/kg/day acrylamide, gavage) and AA+CRV (n:8, 40 mg/kg/day acrylamide, gavage + 50 mg/kg/day carvacrol i.p). At the end of the 15-day study period, the rats, whose blood samples were taken under anesthesia, were euthanized and necropsied. Liver function tests were examined from serums. At the same time for histological analysis, scoring, and hematoxylin-eosin staining, liver tissues were preserved in 10% formaldehyde solution. Results: In microscopic examination, it was determined that AA causes hydropic/vacuolar degeneration, fatty changes, necrosis/apoptosis, bile duct proliferation, hepatic cord dissociation, congestion, megalocytosis and mononuclear cell infiltration. Parallelly, AA raised the levels of ALP, AST, and ALT. The inclusion of CRV to the diet improved histological structure and liver function tests considerably. These results show that CRV has a hepatoprotective effect by reducing AA-induced liver damage. Conclusion: This study showed that CRV attenuated AA-induced liver injury. It is considered that its preventive effect against liver damage is due to its antioxidant properties. However, more research is needed to determine the specific mechanism at the molecular level and offer therapeutic management suggestions.

Detection of bovine coronavirus (BCoV) infection in cattle with clinical respiratory signs by PCR and investigation of the serological status of these animals

Aim: It was aimed to reveal the antigenic and serological prevalence of bovine coronavirus (BCoV) in cattle with clinically respiratory infection and to evaluate the relationship between virus shedding and serological status of animals in this study. Materials and Methods: In the study, nasal swab samples were collected from a total of 92 cattle of different gender between 0 and 2 years of age with clinical respiratory symptoms. These samples were subjected to RT-PCR using specific primer pairs. Furthermore, serum samples were also collected from the same animals and evaluated for BCoV-specific antibodies through an indirect ELISA method. Results: Ten (10.87%) swap samples were defined as positive for BCoV using RT-PCR. The seropositivity rate was 93.48% (86/92) in cattle by ELISA and 90% (9/10) in virus-shedding animals with nasal swab samples. Conclusion: The fact that 93.47% seropositivity was detected in unvaccinated animals against coronavirus in this study indicates that the animals were exposed to BCoV through colostrum or at a certain period of their lives. However the fact that 9 out of 10 animals with BCoV detected in swab samples also have BCoV-specific antibodies casts doubt on the efficacy of these antibodies in protecting against respiratory BCoV infections. We concluded that more detailed studies are needed to reveal the role of antibodies in respiratory coronavirus infections.

A case of plasmacytoma with widespread metastasis in a dog

In this study, a case of plasmacytoma with widespread metastasis in a nine-year-old male Rottweiler dog was described by histopathological and immunohistochemical methods. Macroscopic examination revealed multifocal white or pink-red colored masses ranging in size from 0,5 to 3 cm in the lung, liver, mediastinal and mesenterial lymph nodes, spleen, heart, pancreas, intestine and kidneys. In microscopic examination, tumor foci consisting mostly of atypical plasma cells were observed in the lung, liver, spleen, pancreas, mediastinal and mesenteric lymph nodes, intestine, and kidneys. Tumor-type giant cells were observed in tumor foci in the lung, mitotic figures in tumor foci in other organs, including the lung, and abundant tumor cells in capillary vessels. In Methyl Green Pyronin staining of lung, liver, spleen, pancreas, mediastinal and mesenterial lymph nodes, intestine and kidney sections, the cytoplasm of plasma cells was observed to be stained pink. Congo Red staining applied to liver and spleen sections showed amyloid deposits stained in brick red color. In addition, in the immunohistochemical staining of lung, liver, spleen, pancreas, mediastinal and mesenterial lymph nodes, intestine and kidney sections with Proliferating Cell Nuclear Antigen (PCNA) antibody, severe immunoreactivity was detected in tumor cells. As a result, due to the presence of rarely reported widespread metastases in the lung, liver, spleen, pancreas, mediastinal and mesenterial lymph nodes, intestine and kidneys of the diagnosed plasmacytoma case, it was deemed appropriate to publish the case in order to contribute to the literature by attracting the attention of veterinarians.

Progressive retinal atrophy in Turkish Shepherd Dog; A case report

Progressive retinal atrophy (PRA) is described as an inherited degenerative disease of the retina that is common in dogs. The case was a 9-year-old female Turkish shepherd dog. It was presented to the Small Animal Hospital with complaints of polyuria and stranguria. After examination, the diagnosis was acute hepatitis. Medical treatment (ursofalk, milk thistle, prednisolone and N acetyl cystein) was given. The case lost vision after three days of treatment. The ophthalmological examinations were performed as included slit lamp, Schirmer's tear test, applanation tonometry, fluorescein staining and ultrasound. Ophthalmoscopic and fundoscopic examinations revealed hyperreflectivity and colour change in the tapetal region, marked attenuation of retinal vessels, depigmentation in the non-tapetal region, and optic disc atrophy with scalloped borders. Fundoscopic examination revealed retinal degeneration in both eyes. The aim of this case report is to diagnose PRA in Turkish shepherd dogs, to highlight its significance and to share the results with clinicians.
